Sandwich ELISA - Human Progranulin ELISA Kit (AB252364)
Interpolated concentration of native Progranulin was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 5% human serum. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Sandwich ELISA - Human Progranulin ELISA Kit (AB252364)
Interpolated concentrations of native Progranulin in human milk, saliva, and cell culture supernatant samples.
The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows : milk 1 : 160, saliva 5%, and OVCAR-3 supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 360.2 ng/ml in milk, 60.4 ng/ml in saliva, and 24.4 ng/ml in OVCAR-3 supernatant.

Interpolated concentrations of native Progranulin in human serum and plasma samples.
The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 6.25%, plasma (heparin) 6.25%, and plasma (EDTA) 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 60.9 ng/mL in serum, 47.8 ng/mL in plasma (citrate), 52.2 ng/ml in plasma (heparin), and 49.8 ng/ml in plasma (EDTA).

Product details
Human Progranulin ELISA Kit ab252364 is a rapid single-wash 90-min ELISA kit to measure Human Progranulin in cerebral spinal fluid (CSF), saliva, urine, milk, serum and plasma. This SimpleStep ELISA® sensitivity is 5.64 pg/mL.
How the assay works
Human Progranulin SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
This kit is designed for the quantification of human Progranulin. The standard protein in this kit is mature full-length human Progranulin. Native signal was detected in serum, plasma (heparin), plasma (EDTA), plasma (citrate), cell culture supernatant, milk, saliva, and CSF samples. Spiked protein experiments were used to validate urine and CSF sample types. Cell extract and tissue extract samples have not been tested with this kit.
25% male and female urine samples from healthy donors were measured in duplicate. All values were below the detectable range of the assay.

Biological function summary
Granulin functions as a growth factor involved in tissue repair cell proliferation and inflammation. It does not function as part of a larger protein complex but acts individually in exerting its effects on cells. Granulin influences various aspects of cellular architecture and signaling pathways making it important for maintaining cellular homeostasis. Additionally the granulin protein participates in regulating networks that involve cytokines and growth factors impacting both the innate and adaptive immune responses.
Pathways
Granulin plays significant roles in the MAPK and NF-kB signaling pathways. These pathways critically manage cellular reactions to stress inflammation and other external stimuli. Granulin modulates these processes often interacting with proteins like TNF receptors and interleukins to affect the downstream signaling events. Interactions between granulin and other signaling molecules allow it to integrate into broader networks that oversee various biological responses.
