The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows : milk 1 : 160, saliva 5%, and OVCAR-3 supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 360.2 ng/ml in milk, 60.4 ng/ml in saliva, and 24.4 ng/ml in OVCAR-3 supernatant.

Human Progranulin ELISA Kit ab252364 is a rapid single-wash 90-min ELISA kit to measure Human Progranulin in cerebral spinal fluid (CSF), saliva, urine, milk, serum and plasma. This SimpleStep ELISA® sensitivity is 5.64 pg/mL.

How the assay works
Human Progranulin SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit is designed for the quantification of human Progranulin. The standard protein in this kit is mature full-length human Progranulin. Native signal was detected in serum, plasma (heparin), plasma (EDTA), plasma (citrate), cell culture supernatant, milk, saliva, and CSF samples. Spiked protein experiments were used to validate urine and CSF sample types. Cell extract and tissue extract samples have not been tested with this kit.
25% male and female urine samples from healthy donors were measured in duplicate. All values were below the detectable range of the assay.
