Function
The protein expressed by the GRN gene is a secreted regulator of lysosomal function and a growth factor involved in processes such as inflammation, wound healing, and cell proliferation. It regulates protein trafficking to lysosomes and lysosomal enzyme activity, facilitating lysosomal acidification which leads to the degradation of mature CTSD by CTSB. It also promotes epithelial cell proliferation by blocking TNF-mediated neutrophil activation, thus preventing the release of oxidants and proteases. Additionally, it modulates inflammation in neurons by preserving neuron survival, axonal outgrowth, and neuronal integrity. Granulin-3, -4, and -7 have distinct roles: Granulin-4 promotes proliferation of epithelial cell line A431 in culture; Granulin-3 inhibits epithelial cell proliferation and induces IL-8 secretion; and Granulin-7 stabilizes CTSD by maintaining its aspartic-type peptidase activity through interaction. Human Progranulin ELISA Kit ab252364 is a rapid single-wash 90-min ELISA kit to measure Human Progranulin in cerebral spinal fluid (CSF), saliva, urine, milk, serum and plasma. This SimpleStep ELISA® sensitivity is 5.64 pg/mL.
How the assay works
Human Progranulin SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit is designed for the quantification of human Progranulin. The standard protein in this kit is mature full-length human Progranulin. Native signal was detected in serum, plasma (heparin), plasma (EDTA), plasma (citrate), cell culture supernatant, milk, saliva, and CSF samples. Spiked protein experiments were used to validate urine and CSF sample types. Cell extract and tissue extract samples have not been tested with this kit.
25% male and female urine samples from healthy donors were measured in duplicate. All values were below the detectable range of the assay.

Interpolated concentrations of native Progranulin in human serum and plasma samples.
The concentrations of Progranulin were measured in duplicates, interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 6.25%, plasma (heparin) 6.25%, and plasma (EDTA) 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 60.9 ng/mL in serum, 47.8 ng/mL in plasma (citrate), 52.2 ng/ml in plasma (heparin), and 49.8 ng/ml in plasma (EDTA).

Undiluted samples are as follows: milk 1: 160, saliva 5%, and OVCAR-3 supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Progranulin concentration was determined to be 360.2 ng/ml in milk, 60.4 ng/ml in saliva, and 24.4 ng/ml in OVCAR-3 supernatant.
