Human Proinsulin ELISA Kit (ab242235) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Proinsulin protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Proinsulin with 7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Proinsulin is the precursor of insulin, an essential and primary hormone involved in glucose metabolism. Proinsulin is synthesized in the beta cells of the islets of Langerhans in the pancreas, where it is processed in the ER into active, mature insulin. Active, mature insulin is stored in secretory granules. Rising blood glucose levels lead to the release of active, mature insulin into the blood stream. Proinsulin levels in the blood are elevated with age, in type II diabetes, and during beta cell dysregulation.

Biological function summary
Insulin and proinsulin are indispensable for maintaining glucose homeostasis in the body. Insulin binds to its receptor a receptor tyrosine kinase on target cells like muscle and adipose tissue to facilitate glucose uptake. Proinsulin also has a lesser-known role in insulin regulation and folding. It mostly exists independently rather than being part of a complex with insulin but precursor and mature insulin forms can transiently appear together during synthesis and secretion.
Pathways
Insulin function fits significantly into the signaling pathways such as the insulin signaling pathway and the PI3K/AKT pathway. These pathways importantly involve glucose transporters like GLUT4 and proteins such as IRS-1 that mediate insulin's effects on glucose and lipid metabolism. Insulin signals lead to the translocation of GLUT4 to the plasma membrane enhancing glucose uptake. Therefore insulin is closely related to these critical metabolic pathways.
