Human Proinsulin ELISA Kit (ab242235) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Proinsulin protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Proinsulin with 7 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Proinsulin is the precursor of insulin, an essential and primary hormone involved in glucose metabolism. Proinsulin is synthesized in the beta cells of the islets of Langerhans in the pancreas, where it is processed in the ER into active, mature insulin. Active, mature insulin is stored in secretory granules. Rising blood glucose levels lead to the release of active, mature insulin into the blood stream. Proinsulin levels in the blood are elevated with age, in type II diabetes, and during beta cell dysregulation.

Insulin often referred to as 'insulin' is a peptide hormone with a mass of approximately 5.8 kDa generated by the beta cells in the pancreas. Its precursor proinsulin is synthesized as a larger molecule with a mass of about 9 kDa. Both insulin and proinsulin are primarily expressed in the pancreatic islets of Langerhans. Proinsulin undergoes enzymatic cleavage to form mature insulin and C-peptide. Insulin plays an essential role in glucose metabolism by regulating cellular uptake of glucose.
Biological function summary

Insulin and proinsulin are indispensable for maintaining glucose homeostasis in the body. Insulin binds to its receptor a receptor tyrosine kinase on target cells like muscle and adipose tissue to facilitate glucose uptake. Proinsulin also has a lesser-known role in insulin regulation and folding. It mostly exists independently rather than being part of a complex with insulin but precursor and mature insulin forms can transiently appear together during synthesis and secretion.

Pathways
