Interpolated concentrations of native Prolactin in human pregnant serum and milk.
The concentrations of Prolactin were measured in duplicate and interpolated from the Prolactin standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Prolactin concentration was determined to be 42647 pg/mL in pregnant serum and 1214 pg/mL in milk.

Serum from ten individual healthy human female donors and ten individual healthy male donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Prolactin concentration in females was determined to be 2778 pg/mL with a range of 294 – 18132 pg/mL. The mean Prolactin concentration in males was determined to be 1733 pg/mL with a range of 179 – 7626 pg/mL.

Interpolated concentrations of native Prolactin in human serum and plasma and of spiked Prolactin in cell culture media samples.
The concentrations of Prolactin were measured in duplicates interpolated from the Prolactin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5% plasma (citrate) 100% plasma (heparin) 100% plasma (EDTA) 100% and RPMI media 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Prolactin concentration was determined to be 1066 pg/mL in serum 480 pg/mL in plasma (citrate) 801 pg/mL in plasma (heparin) and 711 pg/mL in plasma (EDTA).

How the assay works
Human Prolactin/PRL ELISA Kit, Chemiluminescent (ab326392) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human Prolactin/PRL (ab326392) protocol summary
1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection
