Human Protein Z SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Protein Z protein in Serum, Cit Plasma, EDTA Plasma, Hep Plasma, Cell culture supernatant, Urine, Saliva, Milk. Quantitate Human Protein Z with 7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips, available in 10-pack (10 x 96-well plates)
-Also available in a fully validated 384-well format.

Example of human Protein Z standard curve in 1X Wash Buffer PT. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentration of native Protein Z was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1:5,000 human serum. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Wash Buffer PT.
Example of human Protein Z standard curve in 1X Wash Buffer PT in 96-well vs. 384-well plate. Background-subtracted data values (mean +/- SD) are graphed.
Serum of ten individual healthy human male donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Protein Z concentration was determined to be 5,437 ng/mL with a range of 3,862 – 10,048 ng/mL.
Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Protein Z concentration was determined to be 3,666 ng/mL with a range of 1,195 – 7,833 ng/mL.
Interpolated concentration of native Protein Z was measured in duplicate at different sample concentrations. Undiluted samples are as follows: urine 25% and saliva 50%. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Wash Buffer PT.
Interpolated concentration of native Protein Z was measured in duplicate at different sample concentrations. Undiluted samples are as follows: Hep G2 cell culture supernatant 25% and milk 1:25. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Wash Buffer PT.
Interpolated concentration of native Protein Z was measured in duplicate at different sample concentrations. Undiluted samples are as follows: serum 1:5,000, plasma (citrate) 1:2,500, plasma (EDTA) 1:2,500, and plasma (heparin) 1:5,000. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Wash Buffer PT.
