Human PYY ELISA Kit (ab255727) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PYY protein in tissue extracts and edta plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human PYY with 8.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Peptide PYY, also known as peptide tyrosine tyrosine, is a short 36- amino acid peptide encoded by the gene *PYY*. PYY is released from L cells in the ileum and colon in response to feeding, and functions by binding to NPY receptors to induce satiety. Two forms of PYY have been identified, PYY1-36 and PYY3-36, with the latter is most common form. PYY is cleaved at the Pro-30 residue by prolyl endopeptidase FAP(Separase) to generate peptide PYY3-36. PYY3-36 binds to the Y2 receptor of the Y family of receptors. PYY inhibits exocrine pancreatic secretion by vasoconstrictor action and inhibits jejunal/colonic mobility, allowing a more efficient digestion and nutrient absorption.

Peptide YY (PYY) also known as PYY1 or PYY3-36 is a 36-amino acid peptide hormone with a molecular weight of approximately 4 kDa. Expressed primarily in the L-cells of the ileum and colon PYY is released into the bloodstream postprandially. This hormone serves as an important messenger in appetite control and energy homeostasis. Peptide YY reduces gastric motility and increases water and electrolyte absorption in the colon playing an integral role in digestion and nutrient absorption processes.
The hormone influences several physiological functions beyond its mechanical roles. Peptide YY hormone function includes inhibiting appetite through interactions with the neuropeptide Y (NPY) receptors in the brain's hypothalamus. It acts together with similar peptides including pancreatic polypeptide (PP) and neuropeptide Y (NPY) establishing a complex regulatory system that governs energy balance and food intake. PYY contributes to short-term and long-term body weight management linking gut to brain signals.
Peptide YY hormone function integrates into the gut-brain axis and the neuroendocrine system. It engages mainly in the regulation of the Y2 receptor pathway interacting with proteins like the ghrelin receptor and insulin pathways. Moreover PYY can influence the feeding pathway by modulating the arcuate nucleus neurons establishing connections with the melanocortin system and agouti-related peptide (AgRP). This integration importantly contributes to satiety and appetite modulation.
Peptide YY is linked to obesity and irritable bowel syndrome (IBS). Altered levels of PYY associate with increased risk of obesity due to dysregulated appetite signals. Disorders like IBS relate to the imbalance of functional gut hormones including PYY which impacts bowel motility and gastrointestinal comfort. Other proteins such as ghrelin and leptin participate in these pathways providing insight into potential therapeutic targets for these conditions.

The PYY standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
The concentrations of PYY were measured in duplicate and interpolated from the PYY standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Undiluted samples are as follows: small intestine extract 37.5 μg/mL and colon extract 150 μg/mL.
