ELISA - Human Rel B ELISA Kit (AB283878)
Interpolated concentrations of native Rel B in human Raji cell extract and Daudi cell extract based on a 75 μg/mL and 150 μg/mL extract load respectively. The concentrations of Rel B were measured in duplicate and interpolated from the Rel B standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Rel B concentration was determined to be 1,016 pg/mL in Raji cell extract and 2,393 pg/mL in Daudi cell extract.

ELISA - Human Rel B ELISA Kit (AB283878)
Example of human Rel B standard curve in 1X Cell Extraction Buffer PTR. The Rel B standard curve was prepared as described in Section 10 of the protocol book. Raw data values are shown in the table in the protocol book. Background-subtracted data values (mean +/- SD) are graphed.

Product details
Human REL B ELISA Kit (ab283878) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of REL B protein in cell extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human REL B with 34.6 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Rel B is a transcription factor belonging to the NF-kB family which plays a crucial role in the maturation of lymph tissue and maturation of mesenchymal cells. While best known for its roles in mediating immune response, this pleiotropic transcription factor also plays a part in the regulation of circadian rhythm, detoxification, and inflammation. Dysregulation of Rel B is a hallmark of many disease states including Rheumatoid Arthritis, immunodeficiency 53, liver fibrosis, lung inflammation, and non-small cell lung cancer. The immunogen from which the antibodies in this kit were raised represent the full-length protein. This protein shares only 86% sequence identity with mouse and rat and is not expected to be highly cross-reactive.

Biological function summary
Rel B serves as a critical component of the alternative NF-κB signaling pathway. It often forms a complex with another NF-κB family member called p52. This complex controls the transcription of genes involved in the immune response cell survival and differentiation. Additionally Rel B contributes to the development of secondary lymphoid organs and modulation of immune responses.
Pathways
Rel B integrates into the non-canonical NF-κB signaling pathway also influencing the canonical pathway to a lesser extent. In the non-canonical pathway Rel B interacts with NIK and p100 to influence immune cell function. This involvement makes Rel B essential in processes like adaptive immunity and inflammatory responses. The protein p52 closely associates with Rel B aiding its activity in these pathways.
