Abcam's anti-Respiratory syncytial virus (RSV) IgA Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgA class antibodies against Respiratory syncytial virus in Human serum and plasma.

A 96-well plate has been precoated with Respiratory syncytial virus antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgA conjugate is added to the wells, which binds to the immobilized Respiratory syncytial virus -specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Respiratory syncytial virus IgA sample captured in plate.

Activity summary

Respiratory Syncytial Virus (RSV) IgA is a type of immunoglobulin A specific to the RSV. This antibody also known as secretory IgA plays a role in mucosal immunity. IgA has two isotopes: IgA1 and IgA2 with IgA2 mostly found in mucosal areas. The molecular weight of IgA is approximately 160 kDa when in its secretory form. Expression occurs predominantly in mucosal surfaces such as respiratory tracts intestines and tears where it acts as the first line of defense against pathogens like RSV.

Biological function summary

RSV IgA acts as an important protector in mucosal areas preventing viral infections at entry points. It is part of the immune system’s humoral response contributing to neutralization and clearance of pathogens. This protein does not work in isolation but forms complexes with the secretory component which shields it from proteolytic degradation and aids in its transport across epithelial layers. Through its protective function RSV IgA reduces the likelihood of systemic spread of viruses by confining them to mucosal surfaces.

Pathways

The involvement of RSV IgA in the mucosal immune response is central to maintaining the host's defense against respiratory pathogens. It participates in the immune exclusion pathway which minimizes viral entry and colonization. RSV IgA interacts with other immunoglobulins such as IgG and IgM working collectively to mount effective immune responses. Additionally it engages in signal transduction associated with the polymeric immunoglobulin receptor (pIgR) facilitating its transcytosis across epithelial cells.

Associated diseases and disorders

RSV IgA is closely linked to respiratory diseases such as bronchiolitis caused by RSV infection. Its presence in sufficient quantity is critical in preventing severe RSV-related lower respiratory tract infections. Insufficient IgA levels can lead to increased susceptibility and severity of these infections. It is also connected to chronic obstructive pulmonary disease (COPD) where impaired mucosal IgA response may contribute to the disease progression. In these contexts RSV IgA works alongside proteins such as pIgR and other components of the immune system to modulate immune responses and control disease outcomes.
