Function

Member of the repulsive guidance molecule (RGM) family that performs several functions in the developing and adult nervous system. Regulates cephalic neural tube closure, inhibits neurite outgrowth and cortical neuron branching, and the formation of mature synapses. Binding to its receptor NEO1/neogenin induces activation of RHOA-ROCK1/Rho-kinase signaling pathway through UNC5B-ARHGEF12/LARG-PTK2/FAK1 cascade, leading to collapse of the neuronal growth cone and neurite outgrowth inhibition. Furthermore, RGMA binding to NEO1/neogenin leads to HRAS inactivation by influencing HRAS-PTK2/FAK1-AKT1 pathway. It also functions as a bone morphogenetic protein (BMP) coreceptor that may signal through SMAD1, SMAD5, and SMAD8.

Human RGMA ELISA Kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human RGMA in serum, plasma and cell culture supernatants. This assay employs an antibody specific for human RGMA coated on a 96-well plate.

Activity summary

Repulsive Guidance Molecule A (RGMa) also known as RGM domain family member A is a glycoprotein involved in axon guidance and neuronal differentiation. It has a molecular weight of approximately 37 kDa. RGMa is expressed broadly in the central nervous system particularly in developing nervous tissues. Beyond the CNS RGMa is also found in various non-neuronal tissues. Its role in axonal repulsion is facilitated by its interaction with the Neogenin receptor influencing the directionality and pathfinding of growing nerve fibers.

Biological function summary

RGMa plays a role in maintaining the structural organization of the nervous system by preventing aberrant nerve growth. RGMa forms part of a larger signaling complex that influences cell differentiation and migration. It acts by binding to Neogenin and recruiting co-receptors such as bone morphogenetic proteins (BMP) which regulate the differentiation and proliferation of neural progenitor cells. This interaction highlights its involvement in maintaining proper neural architecture and functionality.
