Interpolated concentrations of native Ribonuclease 3 in human serum and plasma samples.
The concentrations of Ribonuclease 3 were measured in duplicates, interpolated from the Ribonuclease 3 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 12.5%, plasma (EDTA) 12.5%, and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ribonuclease 3 concentration was determined to be 5,486 pg/mL in serum, 4,124 pg/mL in plasma (citrate), 3,493 pg/mL in plasma (EDTA), and 8,780 pg/mL in plasma (heparin).

Undiluted samples are as follows : PMBC stimulated 12.5% and PBMC unstimulated 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ribonuclease 3 concentration was determined to be 2,787 pg/mL in PBMC stimulated and 1,335 pg/mL in PBMC unstimulated.

How the assay works

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

Ribonuclease 3, also known as Eosinophil Cationic Protein (ECP) is part of the Ribonuclease A family and is able to degrade RNA. Ribonuclease 3 is expressed by the eosinophils and secreted into the blood stream. Serum and plasma levels of Ribonuclease 3 increase in response to inflammatory diseases. Ribonuclease 3 is measured diagnostically to diagnose allergic disorders and other conditions connected with activated eosinophils.
