Interpolated concentrations of native ROBO4 in human cell culture supernatant samples.

The concentrations of ROBO4 were measured in duplicates, interpolated from the ROBO4 standard curves and corrected for sample dilution. Undiluted samples are as follows : HUVEC supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ROBO4 concentration was determined to be 4684.82 pg/mL in HUVEC cell culture supernatant.

The concentrations of ROBO4 were measured in duplicates, interpolated from the ROBO4 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1%, plasma (citrate) 1%, plasma (EDTA) 1% plasma (heparin) 1% and HUVEC supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ROBO4 concentration was determined to be 136.18 ng/mL in serum, 127.46 ng/mL in plasma (citrate), 104.05 ng/mL in plasma (EDTA), and 110.25 ng/mL in plasma (heparin).

0.5 % serum from ten individual healthy human female donors was measured in duplicate.

Human ROBO4 ELISA kit (ab282879) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ROBO4 protein in human serum, plasma, and supernatants. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ROBO4 with 4.56 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Human ROBO4 (Roundabout homolog 4) is a receptor SLIT2 among Slit family of proteins. Human ROBO4 is involved in the maintenance of endothelial barrier organization and function. Studies suggest ROBO4 is involved in angiogenesis and vascular patterning. ROBO4 may mediate the inhibition of primary endothelial cell migration by Slit proteins.

Roundabout homolog 4 (Robo4) is a protein weighing approximately 107 kDa. It belongs to the immunoglobulin superfamily and is sometimes referred to as Magic Roundabout. Robo4 primarily gets expressed in endothelial cells playing an important function in vascular biology. It is characterized by having two immunoglobulin-like domains and two fibronectin type III domains making it distinct among the other Robo family members.
