Human RSPO2 ELISA Kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human RSPO2 in serum, plasma and cell culture supernatants. This assay employs an antibody specific for human RSPO2 coated on a 96-well plate.

RSPO2 also known as R-spondin-2 is a secreted protein weighing approximately 35 kDa. It acts mechanically by potentiating the Wnt signaling pathway. This protein is part of the R-spondin family and contains a signal peptide followed by two furin-like repeat domains a thrombospondin type 1 domain and a C-terminal region. RSPO2 is expressed in various tissues with notable expression in the lung muscle and reproductive organs.
Biological function summary

The protein enhances Wnt signaling which is important for the regulation of cellular processes including proliferation and differentiation. RSPO2 interacts with LGR receptors and is essential in modulating tissue development and homeostasis. The protein forms part of a complex with ZNRF3 triggering removal of negative regulators from the cell surface. This fine regulation allows RSPO2 to play roles in skeletal and organ development.

Pathways

RSPO2 actively participates in the canonical Wnt/β-catenin signaling and non-canonical pathways. Within the Wnt/β-catenin pathway RSPO2 collaborates with LRP6 and Frizzled receptors to stabilize β-catenin which then translocates to the nucleus to influence gene expression. In pathways regulating tissue homeostasis RSPO2 works with RNF43 to control ZNRF3 activity affecting how many receptors are available for Wnt signaling.

RSPO2 shows involvement with craniofacial development disorders and certain cancers. Mutations or dysregulation of RSPO2 and its partners such as LRP6 can lead to craniofacial deformities stemming from improper cell signaling during embryonic development. In cancer RSPO2 may contribute to tumor growth due to its role in enhancing Wnt signaling which is often activated aberrantly in tumor cells. The interplay with ZNRF3 in cancer illustrates its wider impact on tissue overgrowth and malignant transformation.

Activator of the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors. Upon binding to LGR4-6 (LGR4, LGR5 or LGR6), LGR4-6 associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. Also regulates the canonical Wnt/beta-catenin-dependent pathway and non-canonical Wnt signaling by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway (PubMed : 21727895, PubMed : 21909076, PubMed : 22615920). During embryonic development, plays a crucial role in limb specification, amplifying the Wnt signaling pathway independently of LGR4-6 receptors, possibly by acting as a direct antagonistic ligand to RNF43 and ZNRF3, hence governing the number of limbs an embryo should form (PubMed : 29769720).
