Human SEMA4C ELISA kit (ab284623) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human SEMA4C protein in human serum, plasma, cell culture supernatant and cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SEMA4C with 23.5 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. SEMA4C also known as SEMAI is a member of the semaphorin family which are proteins involved in cellular signaling. SEMA4C has a molecular mass of approximately 89 kDa and features in numerous cellular contexts. It is expressed in various tissues including the neural tissue muscle tissue and certain epithelial tissues. This expression suggests a diverse role in different cellular environments contributing to various physiological processes.
SEMA4C plays a significant role in mediating cell-cell communication particularly in processes like axon guidance and cell migration. It acts as a ligand in binding to plexin receptors forming part of signaling complexes that regulate cellular dynamics. This protein can influence cytoskeletal reorganization which is also essential for connections between neurons and the development of the nervous system. SEMA4C is important for both developmental processes and responses in adults where its regulation impacts cellular behavior and function.
SEMA4C modulates signaling cascades such as the MAPK pathway and the PI3K/AKT pathway. These pathways are critical in controlling growth and survival of cells. SEMA4C interacts with other proteins such as Plexin-B1 and Plexin-B2 signaling through these pathways and impacting neuronal development and immune response. It acts as a mediator between the extracellular environment and intracellular signaling mechanisms playing an important role in cellular responses to external stimuli.
SEMA4C is associated with cancer and neurodegenerative diseases. In cancer overexpression of SEMA4C can promote tumor progression and metastasis often linked with other proteins like Plexin-B1 in the tumor microenvironment. In the context of neurodegenerative diseases alterations or dysregulation of SEMA4C signaling can affect neuronal survival and connectivity and it might interact with proteins like tau contributing to disease progression. Understanding the function of SEMA4C in these conditions may provide avenues for therapeutic interventions.

Interpolated concentration of native SEMA4C was measured in duplicate at different sample concentrations. Undiluted samples are 100% serum, plasma-EDTA, plasma-Heparin, and MCF-7 cell culture supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Interpolated concentration of native SEMA4C was measured in duplicate at different sample concentrations. Undiluted samples are 500 μg/mL MCF-7 cell extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.
Interpolated concentrations of SEMA4C in normal human serum donors.
Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +- SD, n=2). The mean SEMA4C concentration was determined to be 301.3 pg/mL with a range of 126.8 – 570.3 pg/mL.
