Sandwich ELISA - Human Semaphorid 4D/CD10 ELISA Kit (AB282868)
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Semaphorin 4D/CD10 concentration was determined to be 12.38 ng/mL with a range of 3.35 – 43.52 ng/mL.

Sandwich ELISA - Human Semaphorid 4D/CD10 ELISA Kit (AB282868)
Interpolated concentrations of native Semaphorin 4D/CD10 in human serum and plasma samples.
The concentrations of Semaphorin 4D/CD10 were measured in duplicates, interpolated from the Semaphorin 4D/CD10 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%, plasma (citrate) 12.5%, plasma (heparin) 25%, and plasma (EDTA) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Semaphorin 4D/CD10 concentration was determined to be 10.88 ng/mL in serum, 14.75 ng/mL in plasma (citrate), 10.21 ng/mL in plasma (heparin), and 11.02 ng/mL in plasma (EDTA).

Product details
Human Semaphorid 4d/CD10 ELISA kit (ab282868) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Semaphorid 4d/CD10 protein in human serum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Semaphorid 4d/CD10 with 22.9 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Semaphorin 4D/CD10 is an R-Ras GTPase-activating protein receptor for PLXNB1 and PLXNB2. Semaphorin 4D is involved in neural development, migration of cerebellar granule cells, and in the immune system. SEMA4D-PLXNB1 interactions are involved in tumor progression through tumor angiogenesis, tumor invasiveness and macrophage regulation.

Biological function summary
CD100 influences various cellular functions by interacting with multiple receptors such as Plexin-B1 and CD72 as part of a larger signaling complex. It contributes to immune response regulation by promoting T-cell activation and B-cell aggregation which are essential for adaptive immunity. In the nervous system it guides neural development including axon growth and synaptic plasticity. CD100's interaction with Plexin-B1 plays a significant role in these developmental processes often affecting cell morphology and movement.
Pathways
Semaphorin 4D is involved in both immune and neural signaling pathways. In the immune system it influences the PI3K-Akt signaling pathway which impacts cell survival and immune cell activation. It associates with pathways involving Plexin-B1 and RhoA to regulate cytoskeletal dynamics affecting cell motility and growth. Additionally CD100's role in neural pathways intersects with proteins like neuropilin-1 influencing semaphorin-mediated signaling essential for neural connectivity.
