Interpolated concentrations of spiked SEMG2 in human serum and female urine samples. The concentrations of SEMG2 were measured in duplicates, interpolated from the SEMG2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, and female urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

The concentrations of SEMG2 were measured in duplicates, interpolated from the SEMG2 standard curves and corrected for sample dilution. Undiluted samples are as follows : male urine 1 : 17 and female urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SEMG2 concentration was determined to be 79.2 μg/mL in male urine and was undetectable in female urine.

Human SEMG2 ELISA Kit (ab240682) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SEMG2 protein in serum and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SEMG2 with 8.4 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
Semenogelin II contributes to the formation of the seminal coagulum which provides an essential function in immobilizing sperm temporarily after ejaculation. This protein interacts within a complex network involving prostate-specific antigen (PSA) which partially degrades SGII to liquefy the coagulum allowing sperm motility and facilitating fertilization. The activity of SGII in semen coagulation and subsequent liquefaction demonstrates its impact on reproductive success.
Pathways
Semenogelin II is integral to the reproductive pathway involving sperm maturation and fertilization. The effort of PSA to degrade SGII and its counterpart Semenogelin I is central to the regulation of semen liquefaction. This interaction is important to the seminal plasma proteolysis pathway. Proteins such as matrix metalloproteinases are also part of this pathway although they have a distinct and less direct relationship with SGII.
