Interpolated concentrations of native SFN in human skin, PC3, and A431 cell extracts. The concentrations of SFN were measured in duplicate and interpolated from the SFN standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

The concentrations of SFN were measured in duplicates, interpolated from the SFN standard curves and corrected for sample dilution. Undiluted samples are as follows : saliva 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SFN concentration was determined to be 120.5 ng/mL in neat saliva.

Human SFN ELISA Kit (ab245711) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SFN protein in edta plasma, saliva, serum, tissue extracts, and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SFN with 115 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Biological function summary
14-3-3 sigma contributes to cell cycle arrest and apoptosis. As part of a complex it interacts with other proteins to ensure proper cell division and prevent abnormal growth. It binds to phosphorylated serine/threonine residues on its target proteins influencing their activity and function. By doing this it exerts control over cell cycle checkpoints and influences DNA damage response mechanisms.
Pathways
14-3-3 sigma interacts with several pathways that are critical for maintaining cell stability and response to stress. It participates in the PI3K/AKT pathway working closely with proteins like AKT1 to facilitate cell survival and growth suppression under stress conditions. Its involvement in this pathway ties it to the regulation of cell proliferation and survival interconnecting with broader cellular networks.
