Sandwich ELISA - Human SHBG ELISA Kit (AB260070)

Interpolated concentrations of native SHBG in human serum and plasma samples. Samples were pooled from healthy 20 male and 20 female volunteers. The concentrations of SHBG were measured in duplicates, interpolated from the SHBG standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 0.2%, plasma (citrate) 0.2%, plasma (heparin) 0.2% and plasma (EDTA) 0.2%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SHBG concentration was determined to be 1466 ng/mL in serum, 1453 ng/mL in plasma (Citrate),1559 ng/mL in plasma (Heparin), and 1348 ng/mL in plasma (EDTA).

Product details

Human SHBG ELISA Kit (ab260070) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SHBG protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human SHBG with 26 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

SHBG, Sex hormone-binding globulin, is a steroid binding protein used to transport dihydrotestosterone, testosterone, and 17-beta-estradiol. It is crucial in the maintenance of plasma steroid hormone levels. This kit targets the mature, full-length, protein form which lacks the N-terminal signaling peptide.

Supplementary information

The protein sex hormone-binding globulin (SHBG) also known as testosterone-estradiol binding globulin (TEBG) plays an important role in binding and transporting sex steroids in the bloodstream. This glycoprotein has a mass of approximately 90 kDa. SHBG is produced mainly in the liver but it is also expressed in other tissues such as the testes ovaries and placenta. It regulates the availability of sex hormones to tissues and influences their bioactivity at the cellular level.
Biological function summary

SHBG regulates the levels of circulating androgens and estrogens by binding tightly to testosterone and estradiol. This binding inhibits the hormones from interacting with their receptors until they are released therefore controlling their action. SHBG can exist as a monomer but often forms a homodimer complex. In addition to binding sex hormones SHBG also interacts with membrane receptors and may participate in the direct signaling pathways that modulate cellular responses.

Pathways

SHBG plays a role in the regulation of the steroid hormone pathway and is related to the transport and metabolic clearance of sex hormones. This pathway involves transformation and transportation of estrogens and androgens affecting genes regulated by these hormones. SHBG interacts with various proteins including albumin which assists in hormone transport and it might influence the androgen receptor pathway altering cell proliferation and differentiation signals.

SHBG levels influence conditions such as polycystic ovary syndrome (PCOS) and type 2 diabetes. In PCOS lower levels of SHBG can result in greater circulating free testosterone which may exacerbate symptoms. In the context of type 2 diabetes alterations in SHBG levels may affect insulin resistance possibly linked with insulin and glucose transport proteins. Understanding the dynamics of SHBG could aid in the development of therapeutic strategies targeting these conditions.

Product protocols

Target data

Functions as an androgen transport protein, but may also be involved in receptor mediated processes. Each dimer binds one molecule of steroid. Specific for 5-alpha-dihydrotestosterone, testosterone, and 17-beta-estradiol. Regulates the plasma metabolic clearance rate of steroid hormones by controlling their plasma concentration.
