Interpolated concentrations of native Slit2 in human serum, plasma and cell culture supernatant samples.
The concentrations of Slit2 were measured in duplicates, interpolated from the Slit2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, plasma (heparin) 25% and PC-3 supernatant 20%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Slit2 concentration was determined to be 349.7 pg/mL in serum, 378.5 pg/mL in plasma (citrate), 261.1 pg/mL in plasma (EDTA), 255.3 in plasma (heparin), and 5,175 pg/mL in PC-3 supernatant.

Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Slit2 concentration was determined to be 212.2 pg/mL with a range of 98.33 – 381.3 pg/mL. Slit2 was undetected in one donor.

Interpolated concentrations of native Slit2 in human PC-3 cell extract sample based on a 200 μg/mL extract load.
The concentrations of Slit2 were measured in duplicate and interpolated from the Slit2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Slit2 concentration was determined to be 1,113 pg/mL in PC-3 cell extract.

Human Slit2 ELISA Kit (ab283885) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Slit2 protein in serum, plasma, cell culture supernatant and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Slit2 with 16.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Slit2, encoded by the SLIT2 gene, is a secreted extracellular matrix glycoprotein. Acts as molecular guidance cue in cellular migration that is classically studied for role in neuronal guidance during development. Slit2 function appear to be mediated by interactions with roundabout homolog receptor and participates in other cellular processes such as proliferation. Studies have shown Slit2 to be involved with angiogenesis, inflammation, and cancer.

Biological function summary
Slit2 functions in guiding axons during neuronal development and preventing them from straying from their pathways. It complexes with Robo receptors to transmit repulsive directional signals. This interaction is essential in the development of the nervous system ensuring proper neural circuit formation and synaptic connectivity. Besides neural guidance Slit2 also regulates other cellular events like angiogenesis and immune cell migration.
Pathways
Slit2 participates in the axon guidance pathway where it influences neural network structuring by interacting with Robo receptors and other guidance cues. The Slit-Robo pathway plays an important role in axonal repulsion working alongside the Netrin signaling pathway that involves proteins like DCC and UNC5. This way Slit2 and its pathways balance attractive and repulsive cues to form functional neuronal networks.
