Interpolated concentrations of native STAT1 in human Hela and MCF7 cell extracts based on a 500 μg/mL and 80 µg/mL extract load respectively.
The concentrations of STAT1 were measured in duplicate and interpolated from the STAT1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean STAT1 concentration was determined to be 35,051 pg/mL in Hela and 31,450 pg/mL in MCF7.

Human STAT1 ELISA Kit (ab253222) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of STAT1 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human STAT1 with 375.5 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Signal transducer and activator of transcription 1, commonly referred as STAT1 is an intracellular transcription factor known to mediate cellular responses to interferons, cytokines, and growth factors. Gene knockout studies have shown that STAT1 plays a key role in the development and function of the immune system as well as cell viability. Phosphorylation of STAT1 protein will promote the dimerization between STAT1 and STAT2 to upregulate and increase expression of interferon stimulated genes. The standard is affinity purified.

Biological function summary
The STAT1 protein influences cell survival and immune responses. It functions as a transcription factor and participates in the interferon signaling pathway. In this setting it forms a complex with other STAT proteins such as STAT2 and STAT3 to regulate gene expression. By binding to specific DNA sequences in the nucleus STAT1 alters transcription in response to extracellular signals. It contributes to antiviral defense mechanisms and antiproliferative activities in various cells.
Pathways
The STAT1 protein plays essential roles in the JAK-STAT signaling pathway and interferon signaling pathway. It operates closely with proteins such as JAK1 and TYK2 which are involved in the phosphorylation process activating STAT1. During this activation STAT1 helps initiate cellular responses to interferons a type of signaling protein released during viral infections. The JAK-STAT pathway facilitates various physiological processes including immune responses and cell growth regulation.
