Interpolated concentrations of native STAT3 in human, mouse, and rat 100 ug/mL HeLa cell extract, 250 ug/mL A431 cell extract, 50 ug/mL mouse NIH3T3 cell extract and 50 ug/mL rat C6 cell extract.
The concentrations of STAT3 were measured in duplicate and interpolated from the STAT3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean STAT3 concentration was determined to be 10,028.97 pg/mL in HeLa cell extract, 3,677.14 pg/mL in A431 cell extract, 3,906.20 pg/mL in mouse NIH3T3 cell extract and 5,866.53 pg/mL in rat C6 cell extract.

How the assay works
Human STAT3 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution

STAT3, Signal transducer and activator of transcription 3, is a transcription factor in the JAK/STAT signaling cascade which responds to cytokines and growth factors. STAT3 has been linked to several processes important for tumorigenesis and diabetes. proteins. The antibodies for this kit were raised against the N-terminal half of the protein which is highly conserved throughout Mammalia. Therefore, this protein has a high chance of cross-reacting with many species.

It functions not only as a transcription factor but also as part of a larger protein complex that operates within the cell nucleus to influence gene expression. This multifaceted role enables STAT3 to control the expression of genes related to cell proliferation survival and differentiation impacting various biological processes. These functions highlight its importance in tissue homeostasis and response to extracellular signals.
Pathways
STAT3 is actively involved in the JAK-STAT signaling pathway a principal route for many cytokines and growth factors and the MAPK pathway. STAT3 interacts with proteins such as JAK kinases which phosphorylate Stat3 and initiate the STAT3 signaling cascade. Additionally it closely associates with the protein Raf1 in the MAPK pathway linking external signals to transcriptional responses. These pathways play a significant role in immune response inflammation and growth signaling.
