Product details

Abcam's TACE Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TACE in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Human TACE coated on a 96-well plate. Standards and samples are pipetted into the wells and TACE present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TACE antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TACE bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimization may be required with urine samples.

The standard is affinity purified.

TACE also known as ADAM17 is a protein that functions as a metalloprotease disintegrin. It has an approximate mass of 100 kDa. TACE cleaves membrane proteins responsible for releasing soluble forms of several important factors such as TNF-alpha. This protein is expressed broadly across various tissues including the heart liver and lungs playing an essential mechanical role in protein shedding.
Biological function summary

TACE facilitates the release of cytokines and growth factors participating in cellular processes like inflammation and cell proliferation. It does not form part of a larger complex but interacts with numerous substrates on the cell surface to regulate their activity by cleaving their ectodomains. This cleavage action modulates signaling pathways influencing cell-cell communication and response to environmental changes.

Pathways

Tissue development immune response and turnover processes often involve TACE activity. It plays a part in the shedding of TNF-alpha which is important in the NF-kB signaling pathway and it is implicated in the MAPK pathway through its effect on EGFR signaling. TACE regulates these pathways by interacting with related proteins like TNFR1 for TNF-alpha signaling and erbB for EGFR modulation.

Overexpression and dysregulated activity of TACE connect this protein to various adverse conditions including cancer and rheumatoid arthritis. In cancer TACE activity promotes tumor progression by increasing growth factor availability working alongside proteins like EGFR. In rheumatoid arthritis excessive shedding of TNF-alpha by TACE increases inflammation interacting with proteins within the inflammatory cascade to exacerbate symptoms.
