Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +- SD, n=2). The mean TARC concentration of detected samples was determined to be 7.28 pg/mL with a range of undetected – 12.6 pg/mL.

Human TARC ELISA Kit (ab289696) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human TARC in serum, plasma (EDTA), plasma (citrate), cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TARC with 1.049 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. TARC also known as Thymus and Activation-Regulated Chemokine or CCL17 is a chemokine with a molecular mass of approximately 8 kDa. TARC is expressed primarily in the thymus and at lower levels in organs like the lung spleen and colon. It functions mechanically as a signaling molecule by binding to its receptor CCR4. This interaction drives the migration and activation of certain immune cells such as T helper 2 (Th2) cells which plays a role in immune response modulation.
Biological function summary

TARC is significant in the immune system's regulation. It is not part of a larger complex but operates independently to direct the movement of immune cells toward sites of inflammation or tissue damage. This chemokine attracts Th2 cells and can influence the balance of T cell responses important for maintaining immune homeostasis and responding to external pathogens. TARC also contributes to the maturation and activation of immune cells having an influential role in inflammatory processes.

Pathways

The TARC-CCR4 axis is integral to the immune response pathway and the inflammatory signaling pathway. Through these pathways TARC interacts with other proteins like CCL22 which also binds to CCR4 working systematically to guide cell movement and modulate immune functions. It ensures that immune responses are directed accurately which is central within different signaling cascades.

TARC is highly relevant to conditions such as atopic dermatitis and asthma. These diseases involve improper immune responses where TARC's role in attracting Th2 cells contributes to the inflammatory environment. In atopic dermatitis TARC levels are often elevated indicating its strong link to disease progression. Connections also exist between TARC and other proteins like IL-4 and IL-13 which are implicated in asthma facilitating a further understanding of the pathogenesis of these conditions.

May be important for the appropriate adaptation of microglial morphology and synaptic plasticity to acute lipopolysaccharide (LPS)-induced neuroinflammation (By similarity). Plays a role in wound healing, mainly by inducing fibroblast migration into the wound (By similarity).
