Function

Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed:21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed:21985311, PubMed:32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.

Human Tau ELISA Kit ab273617 is a rapid single-wash 90-min ELISA kit to measure Human Tau in cerebral spinal fluid (CSF) cell culture extracts, tissue extracts, cell culture media, serum and plasma. This SimpleStep ELISA® sensitivity is 16 pg/mL.

How the assay works

Human Tau SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity

This kit recognizes both native and recombinant human Tau protein in serum, plasma (EDTA and heparin), cell culture supernatant, cerebrospinal fluid, cell and tissue extract samples only. Urine, milk, saliva samples have not been tested with this kit. This kit is incompatible with plasma (citrate) samples.
The kit is 100% reactive with human recombinant Tau-A isoform and 61% reactive with human recombinant Tau-F isoform.
Recombinant human UCHL-1 and alpha synuclein were prepared at 7 ng/mL and assayed for cross reactivity. No cross-reactivity was observed.
Recombinant human UCHL-1 and alpha synuclein were prepared at 50 ng/mL and 2 ng/mL and tested for interference with 2 ng/mL of recombinant human Tau. No interference with was observed.

Tau proteins constitute nine isoforms from a single transcript from the MAPT gene that range from 33-81 kDa. Tau proteins are expressed mainly in the neurons of the central nervous systems. They promote microtubule assembly and stability and might be involved in the establishment and maintenance of neuronal polarity. The C-terminus of Tau binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that Tau functions as a linker protein between both. Hyperphosphorylation of Tau proteins can result in destabilization of microtubule organization, Tau aggregation, and tangle formation. Defective Tau proteins may play a role in diseases of the nervous systems, including Alzheimer disease, Pick disease of the brain, Progressive supranuclear palsy 1 and Parkinson-dementia syndrome. Based on the immunogen design of this ELISA antibody pair, this kit should detect all nine human Tau isoforms and it should have equal affinity towards human, mouse and rat Tau proteins.

    Interpolated concentrations of native Tau in human SH-SY5Y cell treated for 4 hours based on 80 μg/mL extract loads and human brain tissue based on a 3.5 µg/mL extract load.

    The concentrations of Tau were measured in duplicate and interpolated from the Tau standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Tau concentration was determined to be 2,500 pg/mL in SH-SY5Y mock treated extract, 2,233 pg/mL in SH-SY5Y staurosporine treated extract and 1,849 pg/mL in human brain extract. Staurosporine treatment induces Tau cleavage. As expected, there is minimum difference in Tau concentration between the staurosporine treated and mock-treated SH-SY5Y cell extract, as this kit does not differentiate between the full-length and cleaved form of Tau protein. The cleaved Tau fragment can be specifically measured using Cleaved Tau ELISA Kit (Human Asp738/Mouse Asp713) ab269557, Cleaved Tau ELISA Kit (Human Asp738/Mouse Asp713).
