8,800 pg/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT181, Tau pT217, Tau pT231 and Tau pS396) were tested for specificity and for interference with 4,400 pg/mL of recombinant Tau-A ligated with a peptide containing phosphorylated S199 (Tau pS199). The interpolated dilution-corrected values, expressed in % of input (for specificity) and in % of Tau pS199 control (for interference), are plotted (mean +/- SD, n=2).

Interpolated concentration of native Tau (phospho S199) was measured in duplicate at different sample concentrations. Undiluted samples are as follows : brain cortex tissue extract 6 µg/mL, brain hippocampus tissue extract 12 µg/mL, and SH-SY5Y cell extract 16 µg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Human Tau (phospho S199) SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Tau (phospho S199) protein in Cell Lysate, Tissue extracts. Quantitate Human Tau (phospho S199) with 16.3 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Target data

The protein expressed by the MAPT gene promotes microtubule assembly and stability and might be involved in establishing and maintaining neuronal polarity. Its C-terminus binds axonal microtubules, and the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between the two. Axonal polarity is predetermined by MAPT localization within the neuronal cell's domain defined by the centrosome. The short isoforms allow cytoskeleton plasticity, whereas the longer isoforms may preferentially play a role in its stabilization.
