Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Tau (phospho S396) concentration of detected samples was determined to be 125.5 pg/mL with a range of undetected – 125.5 pg/mL.

Recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT181, Tau pT217, Tau pT231 and Tau pS199) were tested for specificity (each at 25,600 pg/mL), and for interference (each at 12,800 pg/mL) combined with 1,600 pg/mL of recombinant Tau-A ligated with a peptide containing phosphorylated S396 (Tau pS396). The interpolated dilution-corrected values, expressed in % of input (for specificity) and in % of Tau pS396 control (for interference), are plotted (mean +/- SD, n=2).

How the assay works

Human Tau (phospho S396) ELISA Kit, Chemiluminescent - Extracellular (ab325894) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed : 21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed : 21985311, PubMed : 32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.
