Interpolated concentrations of native (A) human Tau (phospho T181) and (B) human Tau (total, measured using Human Tau ELISA Kit ab273617) of 6 individual post-mortem human CSF donors were measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Tau (phospho T181) concentration of detected samples was determined to be 405 pg/mL with a range of undetected - 679 pg/mL.

Serum of ten individual healthy human female donors was measured in duplicate using the colorimetric version of this kit (ab325078). Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Tau (phospho T181) concentration of detected samples was determined to be 904 pg/mL with a range of undetected – 904 pg/mL. The nine donor sera with undetected values using the ab325078 had also undetected values using this kit.

Undiluted samples are as follows : pooled post-mortem cerebrospinal fluid 100% and an individual female healthy serum 25%. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid were tested for specificity and interference. 48,000 pg/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT217, Tau pT231, Tau pS199, and Tau pS396) were tested for specificity. Different concentrations of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (24,000 pg/mL : non-phosphorylated Tau, Tau pT231, Tau pS199, and Tau pS396; 6,000 pg/mL : Tau pT217) were tested for interference with 1,500 pg/mL of recombinant Tau-A ligated with a peptide containing phosphorylated T181 (Tau pT181). The interpolated dilution-corrected values, expressed in % of input (for specificity) and in % of Tau pT181 control (for interference), are plotted (mean +/- SD, n=2).

How the assay works

Human Tau (phospho T181) ELISA Kit, Chemiluminescent - Extracellular (ab325895) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed : 21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed : 21985311, PubMed : 32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.
