Interpolated concentration of native Tau (phospho T181) was measured in duplicate at different sample concentrations. Undiluted samples are as follows : pooled post-mortem cerebrospinal fluid 100% and an individual female healthy serum 25%. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Tau (phospho T181) concentration of detected samples was determined to be 904 pg/mL with a range of undetected – 904 pg/mL.

Human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid were tested for specificity and interference. 48,000 pg/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT217, Tau pT231, Tau pS199, and Tau pS396) were tested for specificity. Different concentrations of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (24,000 pg/mL : non-phosphorylated Tau, Tau pT231, Tau pS199, and Tau pS396; 6,000 pg/mL : Tau pT217) were tested for interference with 1,500 pg/mL of recombinant Tau-A ligated with a peptide containing phosphorylated T181 (Tau pT181). The interpolated dilution-corrected values, expressed in % of input (for specificity) and in % of Tau pT181 control (for interference), are plotted (mean +/- SD, n=2).

Target data

The protein expressed by the MAPT gene promotes microtubule assembly and stability and might be involved in establishing and maintaining neuronal polarity. Its C-terminus binds axonal microtubules, and the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between the two. Axonal polarity is predetermined by MAPT localization within the neuronal cell's domain defined by the centrosome. The short isoforms allow cytoskeleton plasticity, whereas the longer isoforms may preferentially play a role in its stabilization.
