Recombinant Tau (phospho T217) was spiked into the following human samples and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are 25% serum, plasma (citrate), plasma (EDTA), and plasma (heparin). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

100 ng/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT181, Tau pS199, Tau pT231 and Tau pS396) were tested for specificity, and for interference with 10 ng/mL of recombinant Tau-A ligated with a peptide containing phosphorylated T217 (Tau pT217). The interpolated dilution-corrected values, expressed in % of input (for specificity) and in % of Tau pT217 control (for interference), are plotted (mean +/- SD, n=2).

How the assay works

Human Tau (phospho T217) ELISA Kit, Chemiluminescent- Extracellular (ab325447) SimpleStep ELISA®  employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time.  By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colormetric ELISAs.

Target data

The protein expressed by the MAPT gene promotes microtubule assembly and stability and might be involved in establishing and maintaining neuronal polarity. Its C-terminus binds axonal microtubules, and the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between the two. Axonal polarity is predetermined by MAPT localization within the neuronal cell's domain defined by the centrosome. The short isoforms allow cytoskeleton plasticity, whereas the longer isoforms may preferentially play a role in its stabilization.
