Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed:21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed:21985311, PubMed:32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.

30 ng/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT181, Tau pS199, Tau pT217 and Tau pS396) were tested for specificity and for interference with 3 ng/mL of recombinant Tau-A ligated with a peptide containing phosphorylated T231 (Tau pT231). The interpolated dilution-corrected values, expressed as % of values of Tau pT231, are plotted (mean +/- SD, n=2).
Interpolated concentration of native Tau (phospho T231) was measured in duplicate at different sample concentrations. Undiluted samples are 50% CSF from a single, post-mortem female donor. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
Recombinant Tau (phospho T231) was spiked into the following samples and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are 25% serum, plasma (citrate), plasma (EDTA), and plasma (heparin). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
Recombinant Tau (phospho T231) was spiked into the following species samples and diluted in a 2-fold dilution series in Sample Diluent NS in 96-well vs. 384-well plates. Undiluted samples are 12.5% serum. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
