Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed:21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed:21985311, PubMed:32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.

Interpolated concentrations of (A) Tau (phospho T231) and (B) total Tau (Human Tau ELISA Kit ab273617) were measured in duplicate at various concentrations of normal human, mouse, and rat brain tissue extract samples. The interpolated dilution-corrected values, normalized per µg extract, are plotted (mean +/- SD, n=2).
Tau (phospho T231) and total Tau (Human Tau ELISA Kit ab273617) were measured in duplicate of lambda phosphatase treated (NEB P0753, 48,000 U/mL for 15 min) and mock treated mouse brain tissue extract samples. The raw O.D. values are plotted (mean +/- SD, n=2).
Interpolated concentrations of (A) human Tau (phospho T231) and (B) human total Tau (Human Tau ELISA Kit ab273617) were measured in duplicate at different sample concentrations of normal and Alzheimer’s disease (AD) post-mortem human brain extract samples. The interpolated dilution factor corrected values, normalized per µg extract, are plotted (mean +/- SD, n=2). Interpolated concentration of native Tau (phospho T231) was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1.5 µg/mL brain cortex tissue extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

30 ng/mL of recombinant human Tau-A proteins each ligated with a peptide containing a specific phosphorylated amino acid (non-phosphorylated Tau, Tau pT181, Tau pS199, Tau pT217 and Tau pS396) were tested for specificity and for interference with 3 ng/mL of recombinant Tau-A ligated with a peptide containing phosphorylated T231 (Tau pT231). The interpolated dilution-corrected values, expressed as % of values of Tau pT231, are plotted (mean +/- SD, n=2).
