The Human TCblR Enzyme-Linked Immunosorbent Assay (ELISA) kit (8D6A) (ab213759) is designed for the quantitative measurement of Human TCblR in cell culture supernatants, serum, plasma (heparin, EDTA) and urine.
The ELISA kit is based on standard sandwich enzyme-linked immunosorbent assay technology. A monoclonal antibody from mouse specific for TCblR has been pre-coated onto 96-well plates. Standards (Expression system for standard: NSO; Immunogen sequence: S36-Y229) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for TCblR is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human TCblR amount of sample captured in plate.
TCblR (cluster of differentiation 320), also known as 8D6A or CD320, is a human gene. It is mapped to chromosome 19p13.2. This gene encodes the transcobalamin receptor that is expressed at the cell surface. It mediates the cellular uptake of transcobalamin bound cobalamin (vitamin B12), and the expression of TCblR enhanced B-cell proliferation and immunoglobulin secretion. Mutations in this gene are associated with methylmalonic aciduria. It has been found that antagonists of the TCblR signaling pathway may counter the growth of follicular lymphomas or other tumors that metastasize to lymphoid follicles. TCblR is also a specific receptor for uptake of TCN2-bound cobalamin.

Biological function summary
TCblR facilitates the cellular uptake of vitamin B12 which is necessary for DNA synthesis and cell growth. It is not part of a larger protein complex but it interacts directly with transcobalamin a carrier protein that escorts vitamin B12 through the bloodstream. The interaction between TCblR and transcobalamin-bound vitamin B12 permits the transport of this vitamin into the cell where it plays a role in metabolic processes.
Pathways
TCblR plays an important role in the cobalamin metabolism pathway by mediating the internalization of vitamin B12 into cells. This uptake is important for the methionine and folate cycles where vitamin B12 functions as a cofactor. TCblR is therefore linked to the methionine synthase enzyme which is part of the folate cycle and necessary for the conversion of homocysteine to methionine.
