Interpolated concentrations of native TFF1 in human serum, plasma and urine samples.
The concentrations of TFF1 were measured in duplicates, interpolated from the TFF1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 2.5%, plasma (citrate) 6.25%, plasma (EDTA) 6.25%, plasma (heparin) 6.25%, and urine 1 : 200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TFF1 concentration was determined to be 18.85 ng/mL in serum, 2.85 ng/mL in plasma (citrate), 2.48 ng/mL in plasma (EDTA), 2.27 ng/mL in plasma (Heparin), and 152.70 ng/mL in urine.

Human TFF1 ELISA kit (ab277704) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human TFF1 protein in human serum, plasma, urine, and cell culture media. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human TFF1 with 2.91 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. TFF1 (Trefoil factor 1), also known as pS2, is a protein that in humans is encoded by the TFF1 gene. This gene and two other related trefoil family member genes are found in a cluster on chromosome 21. Trefoils are stable secretory proteins expressed in gastrointestinal mucosa. TFF1 function is not fully defined but is suggested to protect the gastric mucosa by stabilizing the mucus layer and affect healing of the epithelium. It is found that TFF1 in normal human urine inhibited the growth of calcium oxalate crystals. Urinary TFF1 showed an inhibitory potency similar to nephrocalcin. Concentrations and relative amounts of TFF1 in the urine of patients with idiopathic calcium oxalate kidney stones were significantly less than those found in controls. Mouse and rat share 67 % and 69 % sequence homology to human TFF1.

Biological function summary
This trefoil factor plays a significant role in the process of epithelial regeneration and maintaining gastrointestinal tract integrity. It is not known to be part of a complex but works through interaction with various growth factors and cytokines. Its expression is regulated by estrogen making pS2 significant in estrogen-responsive tissues. These interactions contribute to maintaining the stability of epithelial lining aiding in cellular migration and wound healing.

PS2 influences cellular signaling involved in the gastric mucosal defense mainly interacting with the Epidermal Growth Factor (EGF) pathway. It modulates the pathway's effects on cell proliferation and migration. Its role is linked to the mitigation of tissue damage indicating a connection with other proteins like the EGF receptor contributing to the repair and defense mechanisms of epithelial tissues.
