Human Thioredoxin 1 ELISA Kit (ab277448) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Thioredoxin 1 in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Human Thioredoxin 1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Human Thioredoxin 1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Human Thioredoxin 1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Human Thioredoxin 1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Thioredoxin 1 also known as TRX1 or TXN is a small redox protein with an approximate mass of 12 kDa. Mechanically it functions as a thiol-disulfide oxidoreductase facilitating the reduction of other proteins by cysteine thiol-disulfide exchange. Thioredoxin 1 is ubiquitously expressed found in the cytoplasm and also in the nucleus in response to certain signals. Its expression levels can change depending on cellular conditions highlighting its adaptive role in response to oxidative stress.
Biological function summary

Thioredoxin 1 acts as a central player in cellular processes by maintaining a reduced environment in the cell. It serves as an antioxidant modulating the redox state of proteins and is part of the broader thioredoxin system which includes thioredoxin reductase and NADPH. By promoting the reduction of oxidative species and repairing oxidatively damaged proteins it supports many cellular functions such as cell proliferation and apoptosis. This makes it an important part of the intracellular redox balance.

Pathways

Thioredoxin 1 functions within important signaling cascades including the MAPK and NF-kB pathways. In the MAPK pathway it regulates signal transduction by influencing the phosphorylation status of key proteins while in the NF-kB pathway it affects the activation of transcription factors related to inflammation and immune response. The interaction with these pathways links Thioredoxin 1 to other proteins such as ASK1 and IKK contributing to stress response and inflammation.

Thioredoxin 1 has been implicated in cancer and cardiovascular disease. Its overexpression can lead to persistent reduction states favorable for tumor growth and survival and it's often observed in various cancer types. In cardiovascular disease the dysregulation of its activity contributes to oxidative stress influencing conditions such as atherosclerosis. Through these pathways it connects with proteins like VEGF in cancer and Nitric Oxide Synthase in cardiovascular diseases indicating its diverse role in pathological processes.
