Product details
Abcam's TIE1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TIE1 in cell culture supernatants.
This assay employs an antibody specific for Human TIE1 coated on a 96-well plate. Standards and samples are pipetted into the wells and TIE1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TIE1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TIE1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
We have not been able to detect endogenous Human Tie-1 in normal serum with ab100649, only in serum spiked with Human Tie-1. Plasma is not recommended.
Serum has been validated as a sample matrix with this kit. However, TIE1 concentrations are low in normal serum and therefore may not be detected by this assay.
Plasma is not recommended.
Optimization may be required with urine samples.

Biological function summary
TIE1 associates with angiogenesis and blood vessel integrity. It forms a complex with other proteins notably TIE2 within the endothelial cells where it modulates endothelial cell survival and proliferation. Its role becomes particularly significant in dynamic environments where new blood vessel formation or remodeling occurs. TIE1 interacts with angiopoietins providing a network essential for maintaining vascular homeostasis and integrity.
Pathways
TIE1 interacts within angiopoietin-TIE signaling pathways. This pathway plays a critical role in vascular maturation and stability. TIE1 working in conjunction with TIE2 and the angiopoietins orchestrates the intricate balance between vessel stabilization and sprouting needed for healthy vascular function. These interactions link TIE1 with core processes in endothelial cell communication and vascular development.
