Interpolated concentrations of native TIMP-1 in human serum, plasma, and saliva samples. The concentrations of TIMP-1 were measured in duplicates, interpolated from the TIMP-1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 5%, plasma (EDTA) 5%, plasma (heparin) 10%, and saliva 2%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TIMP-1 concentration was determined to be 95.9 ng/mL in serum, 74 ng/mL in plasma (citrate), 68 ng/mL in plasma (EDTA), 56 ng/mL in plasma (heparin), and 179.4 ng/mL in saliva.

Serum of ten individual healthy human donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean TIMP-1 concentration was determined to be 92.6 ng/mL with a range of 66.5 – 128.7 ng/mL.

The mean TIMP-1 concentration was determined to be 5 ng/mL in unstimulated PBMC cell culture supernatant, 22 ng/mL in stimulated PBMC cell culture supernatant, and undetectable in media.

How the assay works

Human TIMP-1 ELISA Kit, Chemiluminescent (ab325730) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colormetric ELISAs.

Target data

Metalloproteinase inhibitor that functions by forming one to one complexes with target metalloproteinases, such as collagenases, and irreversibly inactivates them by binding to their catalytic zinc cofactor. Acts on MMP1, MMP2, MMP3, MMP7, MMP8, MMP9, MMP10, MMP11, MMP12, MMP13 and MMP16. Does not act on MMP14. Also functions as a growth factor that regulates cell differentiation, migration and cell death and activates cellular signaling cascades via CD63 and ITGB1. Plays a role in integrin signaling. Mediates erythropoiesis in vitro; but, unlike IL3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors.
