Human TIMP2 ELISA Kit ab270213 is a rapid single-wash 90-min Sandwich ELISA to measure Human TIMP2 in cell culture media, cell culture supernatant, citrate plasma, EDTA plasma, heparin plasma, saliva, serum, urine. This SimpleStep sensitivity is 19.5 pg/mL.
How the assay works
Human TIMP2 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

TIMP2 (tissue inhibitor of metalloproteinases 2) is a member of the TIMP gene family that are known to be natural inhibitors of the matrix metalloproteinases. Moreover, TIMP2 has a unique function in its ability to suppress the proliferation of endothelial cells in response to angiogenic factors as well as inhibiting protease activity in tissues undergoing extracellular matrix remodeling. As a result, making TIMP2 critical to the maintenance of tissue homeostasis.
TIMP2 also known as Tissue Inhibitor of Metalloproteinases 2 mechanistically inhibits the activity of matrix metalloproteinases (MMPs). It binds to MMPs regulating the extracellular matrix's remodeling. The protein has a molecular mass of around 21 kDa. TIMP2 is expressed in various tissues including the brain liver and kidneys where it maintains a balance between matrix degradation and production.
TIMP2 plays a significant role in tissue maintenance and repair by controlling extracellular matrix turnover. It forms complexes with MMPs such as MMP-2 inhibiting their proteolytic activities. The protein is also involved in cell proliferation and differentiation affecting processes like angiogenesis and wound healing. Its regulatory function aids in maintaining tissue homeostasis and preventing excessive matrix breakdown.
TIMP2 is integral to the matrix metalloproteinase pathway and the regulation of endothelial cell migration. The protein interacts closely with other MMPs influencing their activation and modulation. Through these pathways TIMP2 holds an important position in cellular migration and signaling ensuring precise matrix remodeling in response to physiological needs.
TIMP2 has significant links with cancer and fibrosis. Dysregulation of TIMP2 expression or its imbalance with MMPs can contribute to tumor progression and metastasis due to uncontrolled matrix degradation. Additionally abnormal TIMP2 levels associate with fibrotic diseases where excessive tissue deposition occurs. TIMP2's interaction with proteins like MMP-9 and MMP-2 highlights its involvement in these pathological conditions.

Human TIMP2 competitor curve comparison.
Standard Curve comparison between human TIMP2 SimpleStep ELISA kit and traditional ELISA kit from leading competitor. SimpleStep ELISA kit shows increased sensitivity.
Example of human TIMP2 standard curve in Sample Diluent NS.
The TIMP2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TIMP2 concentration was determined to be 27,721 pg/mL with a range of 19,755 – 41,782 pg/mL.
Interpolated concentrations of native TIMP2 in human serum and plasma samples.
The concentrations of TIMP2 were measured in duplicates, interpolated from the TIMP2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 15%, plasma (citrate) 15%, plasma (EDTA) 15%, and plasma (heparin) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TIMP2 concentration was determined to be 25,723 pg/mL in serum, 30,263 pg/mL in plasma (citrate), 22,927 pg/mL in plasma (EDTA), and 17,596 pg/mL in plasma (heparin).
Interpolated concentrations of native TIMP2 in human urine and saliva samples.
The concentrations of TIMP2 were measured in duplicates, interpolated from the TIMP2 standard curves and corrected for sample dilution. Undiluted samples are as follows: urine 100%, saliva 12.5%, HepG2 supernatant 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TIMP2 concentration was determined to be 3,088 pg/mL in urine, 5,618 pg/mL in saliva, and 8,400 pg/mL in HepG2 supernatant.
