Human TNF alpha ELISA Kit, Chemiluminescent is a 55 min single-wash SimpleStep Ignite™ ELISA for the rapid quantitation of Human TNF alpha with higher sensitivity and broader measuring range.

Sandwich ELISA - Human TNF alpha ELISA Kit, Chemiluminescent (AB325444)
Interpolated concentrations of native TNF alpha in unstimulated versus stimulated PBMC cell culture supernatant samples. PBMC cells were stimulated with 1.5% PHA-M or vehicle control in 10F RPMI and incubated for 46 hours. The concentrations of TNF alpha were measured in duplicate and interpolated from the TNF alpha standard curves. Undiluted samples are PHA-M stimulated PBMC cell culture supernatant 10% and unstimulated PBMC cell culture supernatant 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TNF alpha concentration was back calculated determined to be 7,265.71 pg/mL in PHA-M stimulated PBMC cell culture supernatant and undetectable in the unstimulated PBMC control.

How the assay works
Human TNF alpha ELISA Kit, Chemiluminescent (ab325444) SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colormetric ELISAs.

Human TNF alpha (ab325444) protocol summary
1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add prepared ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection
