Interpolated concentrations of native Transglutaminase 2 in human plasma (EDTA) sample.
The concentrations of Transglutaminase 2 were measured in duplicates, interpolated from the Transglutaminase 2 standard curves and corrected for sample dilution. Undiluted samples are as follows : plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Transglutaminase 2 concentration was determined to be 1,177 pg/mL in neat plasma (EDTA).

Interpolated concentrations of native Transglutaminase 2 in human HUVEC, K562, HepG2 and A549 cells based on 5 μg/mL, 12 μg/mL, 100 μg/mL and 12 μg/mL extract loads, respectively.
The concentrations of Transglutaminase 2 were measured in duplicate and interpolated from the Transglutaminase 2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Transglutaminase 2 concentration was determined to be 2,780 pg/mL in HUVEC extract, 2,632 pg/mL in K562 extract, 3,236 pg/mL in HepG2 extract and 2,374 pg/mL in A549 extract.

Human Transglutaminase 2 ELISA Kit (ab283882) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Transglutaminase 2 protein in serum, plasma, cell culture supernatant, and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Transglutaminase 2 with 13.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Transglutaminase 2, also known as Protein-Glutamine Gamma-Glutamyltransferase 2, is a calcium-dependent acyltransferase located both intra-cellularly and secreted that catalyzes the formation of covalent bonds between peptide-bound glutamine and various primary amines, such as gamma-amino group of peptide-bound lysine, or mono- and polyamines, thereby producing cross-linked or aminated proteins, respectively. Transglutaminase 2 is involved in many biological processes, such as bone development, angiogenesis, wound healing, cellular differentiation, chromatin modification and apoptosis.
The standard is affinity purified.

Biological function summary
Transglutaminase 2 plays roles in cell adhesion wound healing and the immune response. It mediates extracellular matrix stabilization through crosslinking structural proteins which is critical for tissue repair. The enzyme does not typically form a stable part of a large complex but interacts with fibronectin as a binding partner highlighting its role in cellular adhesion and migration. The enzymatic activity of transglutaminase 2 helps maintain tissue integrity by modifying extracellular and intracellular proteins.
Pathways
Transglutaminase 2 contributes significantly to the integrin-mediated signaling pathway and the apoptosis pathway. It assists in the modulation of cell-matrix interactions by crosslinking matrix proteins which is important for integrin signaling. During apoptosis transglutaminase 2 promotes cellular processes that lead to cell death via its involvement in nuclear condensation and DNA fragmentation. In these pathways proteins such as fibronectin and integrins interact with transglutaminase 2 facilitating its regulatory functions.
