Tryptase is the major neutral protease present in mast cells and is secreted upon the coupled activation-degranulation response of this cell type. May play a role in innate immunity. Isoform 2 cleaves large substrates, such as fibronectin, more efficiently than isoform 1, but seems less efficient toward small substrates (PubMed:18854315).

Interpolated concentration of native Tryptase alpha/beta-1 was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 25% serum. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
Serum of eight individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Tryptase alpha/beta-1 concentration was determined to be 1,994 pg/mL with a range of 553 – 3,713 pg/mL.
Interpolated concentration of native Tryptase alpha/beta-1 was measured in duplicate at different sample concentrations. Undiluted samples are 100% stimulated PBMC supernatant (1.5% PHA-M for 46 hrs) and unstimulated PBMC supernatant (mock treated).
