How the assay works
Human TSLP SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

3. Add TMB development solution - incubate for 10 min

5. Read the results on a plate reader at 450 nm
Thymic stromal lyphopoietin (TSLP) is a protein belonging to the IL-7 cytokine family. TSLP is released by epithelial cells and keratinocytes in response to trauma, infection or allergen. It acts upon dendritic cells to mediate TH2 mediated immune response and appears to be a key mediator in many inflammatory diseases such as: Asthma, Eczema, and Atopic Dermatitis. TSLP also acts upon mast cells promote the release of IL-5, -6, and -13 in the presence of IL-1 and TNF-?.

Biological function summary
TSLP plays a role in mediating immune system responses. It induces the differentiation of T-helper 2 (Th2) cells promoting a specific type of immune response. TSLP does not appear to be part of a larger protein complex but it often functions in association with the TSLPR and IL-7R? receptor complex. It influences the activity and maturation of dendritic cells which are important for initiating immune responses.
Pathways
A mechanical understanding of TSLP reveals its involvement in both the JAK-STAT and NF-kB signaling pathways. It interacts with proteins like JAK2 and STAT5 which are important for the downstream signaling cascade leading to inflammation and immune responses. These pathways play important roles in the immune system's ability to detect and respond to environmental triggers.
