Product details
Human TWEAK ELISA Kit is designed for the quantitative determination of TWEAK in cell culture supernatants, plasma and serum samples.
This assay employs an antibody specific for human TWEAK coated on a 96-well plate. Standards and samples are pipetted into the wells and TWEAK present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-human TWEAK antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TWEAK bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
This ELISA kit shows no cross-reactivity with the following cytokines tested: Human ANGPTL3, beta IG-H3, CA9, Cathepsin B, CD23, CHI3L1, CTLA-4, DKK-4, DPPIV, EDA-A2, Erythropoietin R, FGF-6, FGF-9, Gas 1, GITR, IGFBP-5, IL-1 F5 (FIL1 delta), IL-1 F6 (FIL1 epsilon), IL-1 F7 (FIL1 zeta), IL-1 F8 (FIL1 eta), IL-1 F9 (IL-1 H1), IL-1 F10 (IL-1HY2), IL-1 R5, IL-21 (IL-17C), IL-18, IL-20, IL-34, IL-5 R alpha, IL-10 R alpha, Layilin, Leptin R, Marapsin, Mer, MMP-7, P-Cadherin, Prostasin, PSMA (FOLH1), SIGIRR, TGF beta R3, Coagulation Factor III.

Biological function summary
TWEAK modulates processes like cell proliferation differentiation and apoptosis. This cytokine affects the immune response and tissue regeneration becoming significant in processes that require tissue remodeling. While acting independently TWEAK does not form part of a larger protein complex. Instead it signals mainly through its exclusive receptor Fn14 on responsive cells triggering cellular responses that influence its biological activities.
Pathways
TWEAK plays an important role in the NF-kB and MAPK signaling pathways. These pathways are important for immune response regulation and cell survival. In these pathways TWEAK closely interacts with TNF receptor-associated factors (TRAFs) which facilitate TWEAK signaling. The connections to pathways involving inflammatory response highlight its significance in maintaining homeostasis within the cellular environment.
