UNC5D also known as Unc-5 Netrin Receptor D is a transmembrane protein that is a member of the UNC5 receptor family. It has a molecular mass of approximately 110 kDa. UNC5D is primarily expressed in the human nervous system especially in developing neurons and is also found in tissues like the prostate breast and colon. This receptor serves an important role in cell guidance by interacting with netrin ligands acting as a dependence receptor that triggers apoptosis in the absence of netrin.
Biological function summary

UNC5D functions in axon guidance and neuronal migration. It plays a central role in the regulation of cell survival and apoptosis being part of a receptor complex that includes netrin-1. In the presence of netrin-1 UNC5D helps to mediate cell adhesion and guidance. The absence of its ligand however initiates programmed cell death making its presence significant in maintaining cellular homeostasis. This mechanism has implications in how neural networks are formed and maintained in the body.

Pathways

UNC5D influences both the apoptosis and axon guidance pathways. As a dependence receptor it is deeply involved in controlling cell fate through signaling pathways that govern cell survival and death. The interaction of UNC5D with netrin-1 is key in guiding neural development while also connecting with apoptotic pathways involving proteins such as DCC (Deleted in Colorectal Carcinoma). This relationship signifies its role in linking guidance cue responses with survival signals necessary for proper neural development.

Research implicates UNC5D in certain forms of cancer including prostate cancer and neuroblastoma. In these contexts the expression or suppression of UNC5D influences tumorigenesis. A loss of its pro-apoptotic function can promote cell survival in unwanted growths implicating netrin-1 as a significant interacting protein that disrupts this balance. Additionally altered UNC5D activity may affect neural disorders given its role in nervous system development and maintenance. As such it remains an important target for understanding and potentially treating certain cancers and neural conditions.

Product protocols

Target data

Receptor for the netrin NTN4 that promotes neuronal cell survival (By similarity). Plays a role in cell-cell adhesion and cell guidance. Receptor for netrin involved in cell migration. Plays a role in axon guidance by mediating axon repulsion of neuronal growth cones in the developing nervous system upon ligand binding (By similarity). May play a role in apoptosis in response to DNA damage (PubMed : 24691657). It also acts as a dependence receptor required for apoptosis induction when not associated with netrin ligand (PubMed : 24519068). Mediates cell-cell adhesion via its interaction with FLRT3 on an adjacent cell (By similarity).
