Interpolated concentrations of native uPAR in human serum and plasma samples.

The concentrations of uPAR were measured in duplicates interpolated from the uPAR standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50% plasma (citrate) 50% plasma (EDTA) 50% and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean uPAR concentration was determined to be 5.34 ng/mL in serum 4.30 ng/mL in plasma (citrate) 3.41 ng/mL in plasma (EDTA) and 4.81 ng/mL in plasma (heparin).

Undiluted samples are as follows : urine 10% and stimulated PBMC supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean uPAR concentration was determined to be 12.68 ng/mL in urine and 6.32 ng/mL in stimulated PBMC supernatant.

How the assay works

Human uPAR ELISA Kit, Chemiluminescent (ab326406) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human uPAR (ab326406) protocol summary

Target data

Acts as a receptor for urokinase plasminogen activator (PubMed : 15677461). Plays a role in localizing and promoting plasmin formation. Mediates the proteolysis-independent signal transduction activation effects of U-PA. It is subject to negative-feedback regulation by U-PA which cleaves it into an inactive form.
