Abcam's VCAM1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human VCAM1 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human VCAM1 coated on a 96-well plate. Standards and samples are pipetted into the wells and VCAM1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human VCAM1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of VCAM1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimization may be required with urine samples.

Biological function summary
Adhesion molecules like VCAM1 participate in immune response by recruiting leukocytes to sites of inflammation. VCAM1 helps guide immune cells to directed locations where they perform defense activities. Although VCAM1 is not part of any known large protein complex its interaction with integrins facilitates the migration of immune cells across the endothelium and into tissue.
Pathways
VCAM1 contributes significantly to the leukocyte extravasation process in inflammatory pathways. It plays a role in both the cytokine-cytokine receptor interaction pathway and the NF-kB signaling pathway which are essential for immune response and cell signaling. Through these pathways it interacts with proteins like ICAM1 and various chemokines helping to coordinate the immune cell movement through vascular tissue barriers.
