Human WIF-1 ELISA Kit is designed for the quantitative determination of WIF-1 in cell culture supernatants, plasma and serum samples.

This assay employs an antibody specific for human WIF-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and WIF-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human WIF-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of WIF-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

This ELISA kit shows no cross-reactivity with the following cytokines tested: Human 2B4, ADAM-9, APRIL, BMP-2, BMP-9, Cathepsin L, FABP2, FAP, FGF-19, Galectin-3, HGFR, IFN-alpha/beta R2, IGF-2R, IL-1R6, IL-24, IL-33, Kallikrein-14, Legumain, LOX-1, MBL, Neprilysin, Notch, NOV, Osteoactivin, PD-1, PGRPs, SERPINA4, sFRP-3, Thrombomodulin, TLR2, Transferrin.

Activity summary

WIF1 also known as Wnt inhibitory factor 1 encodes a protein that interacts with Wnt proteins and inhibits their activity. This protein has an approximate molecular mass of 40 kDa and is secreted into the extracellular space. It shows expression in various tissues including the brain lung and kidney. The structure of WIF1 enables it to bind directly to Wnt proteins modulating their signaling capabilities.

WIF1 has been implicated in cancer and osteoarthritis due to its regulatory effect on Wnt signaling. Downregulation of WIF1 often associates with increased Wnt activity contributing to tumor progression in various cancers through interactions with proteins like DKK1 a known Wnt pathway inhibitor. In osteoarthritis altered WIF1 expression levels can affect cartilage degradation by modifying Wnt signaling showing its role in maintaining joint health.
