Human YKL-40 ELISA Kit (ab255719) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of YKL-40 protein in serum, plasma, urine, cell culture supernatant and cerebrospinal fluid. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human YKL-40 with 3.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. YKL-40 also known as chitinase-3-like protein 1 (CHI3L1) is a glycoprotein with a molecular weight of approximately 40 kDa. Scientists often study it for its roles in inflammatory processes. It has expressions in numerous tissues including liver cartilage and the central nervous system. The CHI3L1 protein does not possess chitinase activity unlike other members of the chitinase family yet it interacts with various ligands to influence cellular behavior.
YKL-40 plays significant roles in tissue remodeling and inflammation. It does not form part of a larger complex by itself but influences several cellular processes. This protein affects cell proliferation migration and survival making it involved in both normal physiological processes and pathological conditions. CHI3L1 also exhibits a connection to the immune response where it modulates macrophage activity and interacts with other immune cells.
YKL-40 is integral to the inflammatory and extracellular matrix pathways. It associates with proteins such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) influencing signaling cascades that modulate inflammation. Through these interactions CHI3L1 becomes a mediator of connective tissue metabolism and immune reactions in various pathophysiological states.
YKL-40 shows strong links to conditions like osteoarthritis and certain cancers. In osteoarthritis elevated levels of CHI3L1 reflect increased cartilage degradation while in oncology it correlates with tumor angiogenesis and metastasis potential. Its connection with proteins such as matrix metalloproteinases highlights its role in disease mechanisms highlighting its potential as a biomarker for early detection and disease progression monitoring.

Interpolated concentrations of native YKL-40 in human serum and plasma samples.
The concentrations of YKL-40 were measured in duplicates, interpolated from the YKL-40 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:40, plasma (citrate) 1:20, plasma (heparin) 1:20, and plasma (EDTA) 1:80. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean YKL-40 concentration was determined to be 37.59 ng/mL in neat serum, 16.17 ng/mL in neat plasma (citrate), 18.92 in neat plasma (heparin), and 67.92 ng/mL in neat plasma (EDTA).
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean YKL-40 concentration was determined to be 42.74 ng/mL with a range of 16.87 – 78.32 ng/mL.

The concentrations of YKL-40 were measured in duplicates, interpolated from the YKL-40 standard curves and corrected for sample dilution. Undiluted samples are as follows: urine 100%, HepG2 cell culture supernatant 100%, U-87 MG cell culture supernatant 5%, and CSF 0.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean YKL-40 concentration was determined to be 449.02 pg/mL in neat urine, 123.48 pg/mL in neat HepG2 cell culture supernatant, 18,524 pg/mL in neat U-87 MG cell culture supernatant and 145,710 pg/mL in neat CSF.
