mCherry ELISA Kit ab221829 is a rapid single-wash 90-min ELISA kit to measure Human IL-1 beta in cell culture extracts, cell culture media and cell culture supernatant. This SimpleStep ELISA® sensitivity is 5.64 pg/mL.
How the assay works
Human IL-1 beta SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit is designed for the quantification of mCherry. The standard protein in this kit is full-length mCherry. Spiked protein experiments were used to validate cell culture supernatant and cell extract sample types. Serum, plasma (citrate), plasma (EDTA), plasma (heparin), urine, saliva, milk, CSF, and tissue extract samples have not been tested with this kit.
1,600 pg/mL and 400 pg/mL of recombinant YFP, GFP, and RFP were individually tested for cros reactivity. No cross reactivity was observed.
1,600 pg/mL of recombinant YFP, GFP, and RFP were individually tested for interference with 200 pg/mL of recombinant mCherry. No interference was observed, with an average recovery of 97% of mCherry protein.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature

3. Add TMB development solution - incubate for 10 min

Interpolated concentrations of spiked mCherry in cell culture supernatant samples.
The concentrations of mCherry were measured in duplicates, interpolated from the mCherry standard curves and corrected for sample dilution. Undiluted samples are as follows: RPMI culture media with 10% FBS (100%), and HUVEC supernatant (100%). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean mCherry concentration was determined to be 204.81 pg/mL in spiked RPMI culture media with 10% FBS (100%) and 212.94 pg/mL in spiked HUVEC supernatant (100%). HUVEC cells were cultured in RPMI media with 10% FBS and supernatant collected.

Interpolated concentrations of spiked mCherry in HEK 293T and 143B (wild-type) cell extract samples based on a 200 μg/mL extract load.
The concentrations of mCherry were measured in duplicate and interpolated from the mCherry standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean mCherry concentration was determined to be 182.08 pg/mL in spiked HEK 293T cell extract and 198.36 pg/mL in spiked 143B cell extract.
Raw O.D. values are shown for mCherry protein, YFP, GFP, and RFP.
All proteins were loaded at 400 pg/mL. The average background O.D. is represented by the dashed line. Signal from YFP, GFP, and RFP loaded at 400 pg/mL was equivalent to background.
