Comparison of rhesus macaque and cynomolgus monkey serum, plasma, and PMA + Ionomycin stimulated PBMC cell culture supernatants (24 hours). The concentrations of CCL4 were measured in duplicates, interpolated from the CCL4 standard curves and corrected for sample dilution. The interpolated dilution factor corrected values in neat samples are plotted (mean +/- SD, n=2).

Undiluted samples are as follows : serum neat, plasma (citrate) neat, plasma (heparin) neat, and plasma (EDTA) neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CCL4 concentration was determined to be 140.2 pg/mL in neat serum, 101.6 pg/mL in neat plasma (citrate), 55.1 in neat plasma (heparin), and 109.5 pg/mL in neat plasma (EDTA).

Interpolated concentrations of native CCL4 in rhesus macaque stimulated peripheral blood mononuclear cell (PBMC) culture supernatant samples.
Rhesus macaque PBMCs were cultured in the presence or absence of 50 ng/mL PMA and 1 μg/mL Ionomycin for 24 hours. The concentrations of CCL4 were measured in duplicates, interpolated from the CCL4 standard curves and corrected for sample dilution. Undiluted samples are as follows : stimulate PBMC supernatant 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CCL4 concentration was determined to be 35.89 ng/mL in neat stimulated rhesus macaque PBMC cell culture supernatant and undetectable in unstimulated PBMC cell culture supernatant and naive cell culture media (not shown).

Monkey CCL4 ELISA Kit (ab242229) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CCL4 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Monkey CCL4 with 1.8 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. CCL4 (MIP-1 beta) is a monokine with inflammatory and chemokinetic properties. CCL4 binds to CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant CCL4 induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form CCL4 retains the abilities to induce down-modulation of surface expression of the chemokine receptor CCR5 and to inhibit the CCR5-mediated entry of HIV-1 in T cells. This form of CCL4 is also a ligand for CCR1 and CCR2 isoform B. Structurally, CCL4 forms a heterodimer with MIP-1 alpha and is secreted from peripheral blood lymphocytes.
