Recombinant IL-12 p70 was spiked into the following Monkey samples and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are as follows : Serum 25%, Plasma (Heparin) 25%, Plasma (EDTA) 50%, Plasma (Citrate) 25%, and Cell Culture Media 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

IL12 p70 also known as Interleukin-12 p70 plays a significant role in the immune response. It is a heterodimeric cytokine made up of two subunits p35 and p40 which together form the active p70. The molecular mass of the complete IL12 p70 molecule is approximately 70 kDa. It is primarily produced by activated dendritic cells and macrophages found mostly within the lymphoid tissues and at sites of inflammation.
Biological function summary

IL12 p70 functions to bridge innate and adaptive immunity. It induces the differentiation of naive T cells into Th1 cells an important process for the immune response. This cytokine is part of the IL12 family and functions as a complex with its receptor IL12R. It promotes the production of interferon-gamma (IFN-γ) from T and NK cells enhancing the cytotoxic activities that are important for protection against intracellular pathogens.

Pathways

IL12 p70 is essential in the signaling pathways involved in cell-mediated immunity. It engages the JAK-STAT signaling pathway together with the IFN-γ pathway to activate immune responses against viral and bacterial infections. IL12 p70 interacts with proteins such as STAT4 which is key in transmitting its signal within the immune cells leading to the expression of immune response genes.

IL12 p70 plays an important role in inflammatory diseases and autoimmune conditions. Elevated levels of IL12 p70 are associated with diseases like Crohn's disease where it contributes to chronic inflammation through increased IFN-γ production. It is also implicated in multiple sclerosis a result of its ability to promote Th1 differentiation and inflammation. In these conditions IL12 p70 is often connected with other cytokines like TNF-alpha which collectively exacerbate the inflammatory responses.

Heterodimerizes with IL12B to form the IL-12 cytokine or with EBI3/IL27B to form the IL-35 cytokine. IL-12 is primarily produced by professional antigen-presenting cells (APCs) such as B-cells and dendritic cells (DCs) as well as macrophages and granulocytes and regulates T-cell and natural killer-cell responses, induces the production of interferon-gamma (IFN-gamma), favors the differentiation of T-helper 1 (Th1) cells and is an important link between innate resistance and adaptive immunity. Mechanistically, exerts its biological effects through a receptor composed of IL12R1 and IL12R2 subunits. Binding to the receptor results in the rapid tyrosine phosphorylation of a number of cellular substrates including the JAK family kinases TYK2 and JAK2. In turn, recruited STAT4 gets phosphorylated and translocates to the nucleus where it regulates cytokine/growth factor responsive genes (By similarity). As part of IL-35, plays essential roles in maintaining the immune homeostasis of the liver microenvironment and also functions as an immune-suppressive cytokine (By similarity). Mediates biological events through unconventional receptors composed of IL12RB2 and gp130/IL6ST heterodimers or homodimers. Signaling requires the transcription factors STAT1 and STAT4, which form a unique heterodimer that binds to distinct DNA sites (By similarity).
