Interpolated concentrations of native PD-L1 in cynomolgus serum and stimulated cynomolgus PBMC cell culture supernatant samples.
PBMC cell culture supernatant samples were stimulated with 50 ng/ml PMA and 1 μg/ml ionomycin for 24 hours. The concentrations of PD-L1 were measured in duplicates, interpolated from the PD-L1 standard curves and corrected for sample dilution. Undiluted samples are as follows : cynomolgus serum 50% and stimulated cynomolgus PBMC cell culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L1 concentration was determined to be 2,281.25 pg/ml in cynomolgus serum and 2,546.31 pg/ml in stimulated cynomolgus PBMC cell culture supernatant.

PBMC cell culture supernatant samples were stimulated with 50 ng/ml PMA and 1 μg/ml ionomycin for 24 hours. The concentrations of PD-L1 were measured in duplicates, interpolated from the PD-L1 standard curves and corrected for sample dilution. Undiluted samples are as follows : rhesus plasma (heparin) 50%, rhesus plasma (EDTA) 25%, and stimulated rhesus PBMC cell culture supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L1 concentration was determined to be 860.85 pg/ml in rhesus plasma (heparin), 3,398.46 pg/ml in rhesus plasma (EDTA), and 4,486.45 pg/ml in stimulated rhesus PBMC cell culture supernatant.

Monkey PD-L1 [28-8] ELISA kit (ab279408) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Monkey PD-L1 [28-8] protein in monkey serum, plasma and cell culture supernatant samples. PD-L1 (also known as CD274 or B7-H1) is a membrane bound glycoprotein involved in regulation of the immune system. PD-L1 is expressed on a variety of inflammatory-activated cells as well as some carcinomas and in melanoma. PD-L1 binds to PD-1 and CD80, where it can suppress T cell activation and proliferation as well as induce apoptosis. Levels of PD-L1 are increased in the plasma of cancer patients as well as in cerebrospinal fluid of gliomas. PD-L1 can bind PD-1 in order to regulate T cell apoptosis.

Biological function summary
PD-L1 plays a central role in immune evasion mechanisms utilized by tumors. It is not part of a larger protein complex but directly interacts with PD-1 and CD80. When PD-L1 binds to PD-1 it sends inhibitory signals leading to decreased T cell activation and proliferation allowing cancer cells to avoid immune destruction. PD-L1 expression provides a mechanism for tumors to suppress immune surveillance facilitating tumor progression.
Pathways
PD-L1 is integral to the immune checkpoint pathway which is an important regulator of immune response. The interaction between PD-L1 and PD-1 provides a mechanism for immune tolerance which is part of the broader adaptive immune system pathway. PD-L1 is related to other immune checkpoint proteins such as CTLA-4 in its function to limit autoreactivity and promote immune homeostasis.
