Interpolated concentrations of native Adiponectin in Mouse serum, and plasma samples.
The concentrations of Adiponectin were measured in duplicates, interpolated from the Adiponectin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 666, plasma (EDTA) 1 : 666, plasma (heparin) 1 : 666, and plasma (citrate) 1 : 666. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Adiponectin concentration was determined to be 2212 ng/mL in serum, 2168 ng/mL in plasma (EDTA), 2129 ng/mL in plasma (heparin) and 1845 ng/mL in plasma (citrate).

How the assay works
Mouse Adiponectin SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

Adiponectin is a 30kDa monomer that combines to form trimers (low molecular weight), hexamers (middle molecular weight), and other higher molecular weight multimers. Adiponectin is an important adipokine involved in fat metabolism and insulin sensitivity. Insulin stimulates the secretion of adiponectin. Decreased levels of adiponectin are associated with insulin resistance, whereas high levels of adiponectin are associated with reduced risk of type-2 diabetes.

Biological function summary
Adiponectin influences glucose regulation and fatty acid oxidation. It acts as a hormone with several metabolic roles including anti-diabetic anti-atherogenic and anti-inflammatory properties. Adiponectin participates in forming a complex with other proteins such as AdipoR1 and AdipoR2 which are receptors facilitating signal transduction. The interaction of adiponectin with its receptors leads to the induction of several lipid and glucose metabolism pathways important for maintaining cellular energy balance.
Pathways
Many regulatory cascades are influenced by adiponectin. This protein is integral to the AMPK signaling pathway and the PPAR signaling pathway. In the AMPK pathway adiponectin enhances insulin sensitivity and stimulates oxidation of fatty acids in muscle tissue. Through the PPAR pathway it influences lipid metabolism and storage. Adiponectin interacts with key proteins such as leptin and resistin coordinating various metabolic pathways to balance energy and glucose levels making it a critical factor in metabolic regulation.
